Zafgen Inc., a Cambridge, Mass.-based biopharma company focused on the needs of severely obese patients, has raised $21 million in Series D funding. Alta Partners led the round, and was joined by return backers Atlas Venture and Third Rock Ventures. www.zafgen.com

Flexion Therapeutics Inc., a Woburn, Mass.-based developer of therapeutics for musculoskeletal disorders, has raised $20 million in Series B funding. Novo Ventures led the round, and was joined by return backers 5AM Ventures, Pfizer Venture Investments, Sofinnova Partners and Versant Ventures.www.flexiontherapeutics.com

Poshmark, a Menlo Park, Calif.-based mobile marketplace for fashion, has raised $12 million in Series B funding. Menlo Ventures led the round, and was joined by return backers Mayfield Fund, Inventus Capital, and SoftTech VC. The company previously raised $3.5 million. www.poshmark.com

ScaleIO, a Palo Alto, Calif.-based software-defined storage startup, has raised $12 million in VC funding from Greylock IL Partners, Norwest Venture Partners and individual angels. www.scaleio.com

Adelphic Mobile, a Boston-based mobile advertising platform, has raised $10 million in VC funding.Google Ventures led the round, and was joined by return backer Matrix Partners. www.adelphic.com

Knoa Software, a New York-based provider of user experience and performance management software, has raised $7.5 million in new VC funding. Ascent Ventures and FA Tech were joined by return backers Gefinor Ventures, Advantage Capital Partners and Rand Capital. www.knoa.com

QuBit, a London-based customer data platform, has raised $7.5 million in Series A funding led byBalderton Capital. www.qubitproducts.com

Green Throttle Games, a Santa Clara, Calif.-based video game publisher and maker of a new wireless game controller, has raised $6 million in Series A funding. Trinity Ventures led the round, and was joined by DCM. Green Throttle was co-founded by Charles Huang, co-creator of Guitar Hero.www.greenthrottle.com

Magenta Medical, an Israeli medical device startup focused on congestive heart failure, has raised $4 million in seed funding from Pitango Venture Capital. www.pitango.com

SmartThings, developer of a platform for connecting the Internet to everyday items, has raised $3 million in seed funding. First Round Capital led the round, and was joined by SV Angel, Lerer Ventures and CrunchFund. www.smartthings.com

Cadens Imaging, a Québec -based developer of visualization solutions in computer-assisted medical imaging, has raised C$2.7 million in seed funding. Desjardins-Innovatech led the round, and was joined by FSIT, Anges Québec, FIER Montestrie Capital, FIER Croissance Durable, FIER Innovation Durable and FIER Succès. www.cadensimaging.com

CapLinked, a Los Angeles-based provider of a cloud-based platform for business transactions, has raised $1.6 million in Series A funding. Backers include FF Angel, Siemer Ventures, 500 Startups and Peter Thiel. www.caplinked.com

Red Tricycle, an email newsletter that provides parents with local kids activities, has raised $1.5 million in Series A funding. Maveron led the round, and was joined by 500 Startups, Richmond Park Partners, Zulily and individual angels. www.redtri.com

Radio Runt, a Seattle-based developer of a group-centric audio communications application for social and interest networks, has raised $1 million in seed funding from Kenges Rakishev. www.rall.ee
